In calculus, we have to find the slope of a curved line. So we use a secant line to figure it out. We have two points, one will be (x, f(x)) and the other will be (x+h, f(x+h)) like in the picture above. We use the slope formula, (y2-y1)/(x2-x1), which is rise over run. We plug the points into the formula and will get f(x+h)-f(x)/-x+h+x. The x's will cancel out and we will end with the difference quotient.
